Mélange d'action, de stratégie et de jeu de rôle, Lock's Quest sur DS reprend le concept des Tower Defense pour en faire un jeu original et addictif. Le joueur devra construire des fortifications et assumer la défense de son territoire face à des vagues successives de monstres. Un mode multijoueur permet en outre à deux adversaires de s'affronter à coups de stylet.
