Pilotwings 64 est un jeu de simulateur de vol sur Nintendo 64. Comme dans son prédécesseur, le joueur doit remplir une série de missions en utilisant différents véhicules aériens (jetpack, hélicoptère, deltaplane, parachute...). Les points gagnés dans chaque mission sont attribués en fonction du temps, des dommages, du carburant, de la précision, de l'atterrissage et quelques autres critères.
